Lung Expression of Human Angiotensin-Converting Enzyme 2 Sensitizes the Mouse to SARS-CoV-2 Infection

Kun Han et al.

Summary: A rapidly deployable COVID-19 mouse model was developed and characterized by transducing hACE2-expressing mice with recombinant adenovirus and infecting them with SARS-CoV-2, resulting in a mild COVID-19 phenotype. Transcriptomic analysis revealed upregulation of IFN-dependent chemokines and genes associated with effector T-cell populations, demonstrating a correlative response to human COVID-19 cases. This model is useful for preclinical and pathogenesis studies of COVID-19.

The potential significance of high avidity immunoglobulin G (IgG) for protective immunity towards SARS-CoV-2

Georg Bauer

Summary: Avidity plays a crucial role in determining the strength of binding between IgG and its target epitope. The lack of high avidity IgG may lead to insufficient protective immunity. It is suggested to include avidity determination in evaluating vaccination protocols for SARS-CoV-2 to achieve truly protective immunity.

mRNA vaccine-elicited antibodies to SARS-CoV-2 and circulating variants

Zijun Wang et al.

Summary: Volunteers who received the Moderna or Pfizer-BioNTech vaccine showed high levels of antibodies and memory B cell responses against SARS-CoV-2, with activity similar to individuals who had recovered from natural infection. However, their efficacy against specific SARS-CoV-2 variants was reduced, indicating a potential need for periodic updates to mRNA vaccines to maintain clinical efficacy.

Neutralization of SARS-CoV-2 lineage B.1.1.7 pseudovirus by BNT162b2 vaccine-elicited human sera

Alexander Muik et al.

Summary: A new SARS-CoV-2 lineage B.1.1.7 has emerged in the UK, which is more transmissible and faster spreading than other strains. However, a study found that the BNT162b2 vaccine offers largely preserved protection against the B.1.1.7 lineage, despite some reduced neutralizing titers.

Immunogenicity of prime-boost protein subunit vaccine strategies against SARS-CoV-2 in mice and macaques

Hyon-Xhi Tan et al.

Summary: The study compared the immunogenicity of spike proteins (S) and receptor binding domain (RBD) in SARS-CoV-2 vaccines in mice and macaques, finding that S was highly immunogenic in mice while RBD was comparatively poor. Both S and RBD vaccines were similarly immunogenic in macaques, inducing serological neutralising activity exceeding convalescent human levels.

Directed evolution of potent neutralizing nanobodies against SARS-CoV-2 using CDR-swapping mutagenesis

Jennifer M. Zupancic et al.

Summary: There is a strong interest in developing effective methods for generating neutralizing antibodies and nanobodies against SARS-CoV-2. This study presents a simple directed evolution method for producing nanobodies with high affinities and neutralization activities, which can effectively neutralize both SARS-CoV-2 pseudovirus and live virus.
