Whole-genome sequencing to detect mutations associated with resistance to insecticides and Bt proteins inSpodoptera frugiperda

Fang Guan et al.

Summary: The study identified three amino acid substitutions in the ace-1 gene of S. frugiperda associated with organophosphate resistance, with the Brazilian population having all three mutations and the 227A allele being the most common. Populations from China, Malawi, and Uganda carried two of the three ace-1 mutations, with the 290V allele as the dominant allele.

Development of a loop-mediated isothermal amplification (LAMP) assay for the identification of the invasive wood borer Aromia bungii (Coleoptera: Cerambycidae) from frass

Domenico Rizzo et al.

Summary: The red-necked longhorn beetle Aromia bungii is a major pest of cultivated and ornamental species of the genus Prunus in East Asia. While traditional methods require morphological or molecular discrimination to identify this wood borer, non-invasive molecular diagnostic tools can be applied to biological traces for detection without causing extensive plant damage. The study presents a protocol using real-time and visual loop-mediated isothermal amplification for detecting A. bungii DNA extracted from larval frass, demonstrating robustness and high sensitivity in laboratory validations. The visual LAMP protocol could be a useful tool for rapid monitoring and outbreak management of A. bungii.

Geographic Monitoring of Insecticide Resistance Mutations in Native and Invasive Populations of the Fall Armyworm

Sudeeptha Yainna et al.

Summary: This study investigated the resistance of fall armyworm populations to Bt and synthetic insecticides in different geographic areas, finding that resistance mutations were more widespread in invasive populations, resulting in susceptibility to Bt insecticides and resistance to synthetic insecticides in Chinese populations. This highlights the importance of monitoring insecticide resistance in invasive populations using genomic approaches and bioassay experiments.

Summary: Fall armyworm is an economically important pest worldwide and has been identified in Australia. A sensitive, specific, and in-field capable loop-mediated isothermal amplification (LAMP) assay has been developed for rapid and accurate detection of S. frugiperda larvae, which can greatly assist with timely management decisions.

A novel reference dated phylogeny for the genus Spodoptera Guen′ee (Lepidoptera: Noctuidae: Noctuinae): new insights into the evolution of a pest-rich genus

Gael J. Kergoat et al.

Summary: The genus Spodoptera consists of 31 species with varied host plant breadths, with some species being major economic pests. Comparative genomics studies have highlighted major adaptive changes in genetic architecture among different Spodoptera species. Recent population genomics studies provide insights into population structures, migration patterns, and invasion processes of two species.

A LAMP (loop-mediated isothermal amplification) test for rapid identification of Khapra beetle (Trogoderma granarium)

Lea Rako et al.

Summary: The novel Khapra and 18S LAMP assays are able to amplify target beetle DNA quickly and sensitively, even from partially degraded samples, improving speed, accuracy, and confidence of detection at incursion points. The assays are portable and easy to implement in field conditions with limited resources, aiding rapid biosecurity responses in countries affected by Khapra beetle infestations.

Corn-strain or rice-strain? Detection of fall armyworm, Spodoptera frugiperda (JE Smith) (Lepidoptera: Noctuidae), in northern Australia

Maxine P. Piggott et al.

Summary: The fall armyworm is a noctuid moth from the Americas that poses a threat to a wide range of important crops. It consists of two strains with different host plant preferences. While the majority of specimens in Australia belong to the 'rice-strain', there are also 'corn-strain' individuals. Genetic characterization and monitoring of host plant preferences will provide valuable information as the pest spreads to new locations in Australia.
