Clinical outcomes in T-cell large granular lymphocytic leukaemia: prognostic factors and treatment response

Zachary Braunstein et al.

Summary: T-cell large granular lymphocytic leukemia (T-LGLL) is an incurable disease characterized by abnormal cytotoxic T cell proliferation. Clinical outcomes and future treatment directions were evaluated in 60 patients, with cyclophosphamide showing potential as a second-line therapy. Studies on novel agents are needed for patients with relapsed or refractory T-LGLL.

Single-Antibody Evaluation of T-Cell Receptor β Constant Chain Monotypia by Flow Cytometry Facilitates the Diagnosis of T-Cell Large Granular Lymphocytic Leukemia

Pedro Horna et al.

Summary: The addition of TRBC1 antibody to the routine flow cytometry panel can aid in the diagnosis of T-LGLL, simplifying the diagnostic process. Analysis of T-LGLL and reactive lymphocytoses revealed the potential diagnostic value of TRBC1 in assessing cell clonality.

Large granular lymphocytic leukemia - A retrospective study of 319 cases

Summary: Large granular lymphocytic leukemia (LGLL) is a rare hematological malignancy arising from cytotoxic T or NK cells, often accompanied by autoimmune diseases and coexisting malignancies, with a high treatment need. Methotrexate, cyclophosphamide, and cyclosporine A are commonly used drugs with varying response rates, influenced by factors such as thrombocytopenia and autoimmune diseases.

Clinicobiological Characteristics and Outcomes of Patients with T-Cell Large Granular Lymphocytic Leukemia and Chronic Lymphoproliferative Disorder of Natural Killer Cells from a Single Institution

Andrea Rivero et al.

Summary: T-LGLL and CLPD-NK are infrequent lymphoproliferative syndromes characterized by clonal expansions of cytotoxic T lymphocytes and NK cells, with a proportion of patients harboring STAT3 mutations. Patients typically present with anemia and neutropenia, sometimes accompanied by autoimmune conditions and solid tumors. Survival rates of these patients do not differ from the general Spanish population, indicating the indolent nature of these diseases.
