An overview of PLGA in-situ forming implants based on solvent exchange technique: effect of formulation components and characterization

Tarek Metwally Ibrahim et al.

Summary: Due to low oral bioavailability of certain drugs, there is growing interest in parenteral administration to directly target drug absorption into the blood. Injectable in-situ forming implants (ISFIs) are gaining popularity compared to other sustained release systems, offering easier production, acceptable administration routes, reduced dosing frequency, and improved patient compliance. The use of biodegradable polymers like PLGA in ISFI systems allows for extended drug delivery through solvent exchange mechanisms, with drug release characterized by initial burst, diffusion processes, and polymeric degradation. Suitable solvent selection and consideration of PLGA hydrophobicity and molecular weights are crucial for optimizing drug release in ISFIs.

Electrospun Nanofibers of Polycaprolactone/Collagen as a Sustained-Release Drug Delivery System for Artemisinin

Peipei Huo et al.

Summary: In this study, ART-loaded polycaprolactone (PCL)/collagen (Col) nanofibers with different proportions of polymers were prepared and characterized, with a focus on ART anti-crystallization and release behaviors. Various analytical techniques confirmed the differences in performance of ART-loaded PCL/Col nanofibers due to different polymer ratios, with sustained drug release for up to 48 hours following the Fickian release mechanism.

Brain targeting efficiency of intranasal clozapine-loaded mixed micelles following radio labeling with Technetium-99m

Sinar Sayed et al.

Summary: The study aimed to improve the central systemic bioavailability of CLZ by designing polymeric nanomicellar systems targeted to the brain via intranasal delivery. Optimization was based on factors such as transmittance percentage, solubilization efficiency, and particle size, resulting in an optimized formula with small size, high transmittance percentage, and effective solubilization. In vitro and in vivo experiments showed promising results in enhancing the bioavailability of CLZ in the central nervous system through intranasal delivery of optimized formula M5.
