Hemorrhagic Transformation After Tissue Plasminogen Activator Treatment in Acute Ischemic Stroke

Chengli Liu et al.

Summary: This article reviews the recent research progress on hemorrhagic transformation (HT) in ischemic stroke patients treated with thrombolysis. It discusses the mechanisms, experimental models, and biomarkers of HT, as well as the limitations of different models. Furthermore, it investigates the molecular mechanism and clinical features of HT, and outlines neuroprotective strategies to improve HT. The authors suggest further efforts to reduce the risk of HT and improve the clinical prognosis of patients treated with thrombolysis.

Targeting the CD146/Galectin-9 axis protects the integrity of the blood-brain barrier in experimental cerebral malaria

Hongxia Duan et al.

Summary: The study found that endothelial CD146 plays a crucial role in experimental cerebral malaria, with its overexpression leading to the aggregation of infected red blood cells and lymphocytes, thereby disrupting BBB integrity. Deletion of endothelial CD146 or treatment with anti-CD146 antibody can prevent severe symptoms of cerebral malaria, and combined treatment with anti-CD146 antibody and antiparasitic drugs can improve cognition and memory in patients with cerebral malaria.

IL-17 stimulates neutrophils to release S100A8/A9 to promote lung epithelial cell apoptosis in Mycoplasma pneumoniae-induced pneumonia in children

Suwen Bai et al.

Summary: Mycoplasma pneumoniae-induced pneumonia is a common cause of respiratory tract infections in children, but diagnosing it early is challenging due to a lack of good diagnostic methods. Research found that S100A8/A9 protein was highly expressed in children with MPP and could potentially serve as a new biomarker for clinical diagnosis.

Ethanol Gestational Exposure Impairs Vascular Development and Endothelial Potential to Control BBB-Associated Astrocyte Function in the Developing Cerebral Cortex

Michele Siqueira et al.

Summary: Ethanol exposure can lead to permanent damage to the development of the fetal central nervous system, characterized by increased vessel density, aberrant distribution of perivascular astrocytic endfeet, and disrupted endothelial cell function, ultimately affecting blood-brain barrier establishment.

Sphingosine-1-phosphate receptor 3 is implicated in BBB injury via the CCL2-CCR2 axis following acute intracerebral hemorrhage

Dingkang Xu et al.

Summary: The levels of S1PR3 and its ligand, S1P, were significantly increased after ICH, regulating the expression of CCL2 and p38MAPK. Administration of the S1PR3 antagonist CAY10444 to rats resulted in reduced brain edema, improved BBB integrity, and behavioral deficits. Furthermore, increased expression of CCL2, p-p38MAPK, and ICAM-1, as well as decreased ZO-1 expression, was observed after S1P stimulation in cocultured human astrocytes and hCMEC/D3 cells. Inhibition of S1PR3 led to decreased expression of CCL2, p-p38 MAPK, and ICAM-1, and increased ZO-1 expression. Additionally, microglial proliferation and M1 polarization were attenuated after CAY10444 administration.

miR-155 and miR-146a collectively regulate meningitic Escherichia coli infection-mediated neuroinflammatory responses

Bo Yang et al.

Summary: E. coli infection induced upregulation of miR-155 and miR-146a in astrocytes, which modulated bacteria-triggered neuroinflammatory responses through negative feedback regulation involving the TLR-mediated NF-kappa B and EGFR-NF-kappa B signaling pathways, thus protecting the central nervous system from further neuroinflammatory damage.

LncRSPH9-4 Facilitates Meningitic Escherichia coli-Caused Blood-Brain Barrier Disruption via miR-17-5p/MMP3 Axis

Bojie Xu et al.

Summary: This study revealed that in bacterial meningitis infection, overexpression of lncRSPH9-4 competitively sponges miR-17-5p, increasing MMP3 expression, leading to disruption of tight junctions in hBMECs.

Meningitic Escherichia coli α-hemolysin aggravates blood-brain barrier disruption via targeting TGFβ1-triggered hedgehog signaling

Jiyang Fu et al.

Summary: Infection with E. coli can disrupt the blood-brain barrier by attenuating intercellular communication within the barrier, and activating hedgehog signaling could potentially be a protective strategy for future therapy of bacterial meningitis.

Role of Intermediate Filaments in Blood-Brain Barrier in Health and Disease

Ece Bayir et al.

Summary: The blood-brain barrier is a highly selective cellular monolayer unique to the central nervous system microvasculature, mediating communication with the body by regulating the passage of molecules. Recent studies have highlighted the importance of intermediate filaments in the formation and function of cell-cell junctions, particularly vimentin in endothelial cells. Intermediate filaments may have been overlooked as key targets in regulating blood-brain barrier permeability in health and disease.
