Inhibitor-Mediated Structural Transition in a Minimal Amyloid Model

Priyadarshi Chakraborty et al.

Summary: This study presents the crystal structure of a typical amyloid-forming minimal dipeptide, showing a canonical beta-sheet structure at the atomic level. Interestingly, in the presence of an inhibitor, the supramolecular packing of the dipeptide molecules rearranged into a supramolecular 2-fold helix. Overall, this research contributes to our understanding of the mechanism of amyloid formation and the structural transitions that occur during the inhibition process in a fundamental model.

Summary: Research has shown that in systems of self-replicating molecules, simple and fast replicators can be replaced by more complex and slower ones, with the latter being more proficient in catalysis. The use of chemical fueling can maintain replicator systems out of equilibrium, leading to the emergence of more complex replicators that enable open-ended evolution.

Bioactive scaffolds with enhanced supramolecular motion promote recovery from spinal cord injury

Z. Alvarez et al.

Summary: Researchers have successfully utilized peptide amphiphile supramolecular polymers containing two distinct signals to achieve cell signaling and tissue regeneration in a mouse model of severe spinal cord injury. By mutating the peptide sequence of the monomers, they intensified the motions of molecules within scaffold fibrils, resulting in notable effects such as vascular growth, axonal regeneration, and functional recovery. The study suggests that optimizing cell signaling through ensembles of molecules could be achieved by tuning their internal motions.

Unveiling Polymerization Mechanism in pH-regulated Supramolecular Fibers in Aqueous Media

Nicolas M. Casellas et al.

Summary: In this study, a designed and synthesized amine functionalized C-3-symmetric benzotrithiophene (BTT) monomer was used to form pH responsive one-dimensional supramolecular polymers in aqueous media. Instead of simply looking at the effect of pH on aggregate size, a detailed mechanistic study was carried out, revealing a dramatic and reversible change in polymerization mechanism and supramolecular fiber size by modifying pH to induce formation of positively charged ammonium groups. This change was attributed to Coulombic repulsive forces and higher monomer solubility, and was further utilized to control one-dimensional supramolecular polymerization and copolymerization processes through induced frustrated growth of fibers.
