The key role of Warburg effect in SARS-CoV-2 replication and associated inflammatory response

Philippe Icard et al.

Summary: The current mortality due to Covid-19 pandemic highlights the lack of effective treatment, with research suggesting that the replication of SARS-CoV-2 may rely on altered glucose metabolism similar to the Warburg effect in cancer. Inhibiting two key signaling pathways that sustain aerobic glycolysis could potentially inhibit virus replication. Understanding the role of the Warburg effect in Covid-19 could be crucial in developing strategies to reduce infectivity and immune response.

A metabolic modeling approach reveals promising therapeutic targets and antiviral drugs to combat COVID-19

Fernando Santos-Beneit et al.

Summary: Using Flux Balance Analysis, key human metabolic enzymes for therapeutic intervention against COVID-19 were identified, with potential inhibitors predicted. Promising molecules such as Triacsin C and Celgosivir showed effectiveness against various viruses. Other drugs targeting lipid metabolism, carbohydrate metabolism, or protein palmitoylation were also proposed.

Inhibition of Phosphatidylinositol 3-Kinase by Pictilisib Blocks Influenza Virus Propagation in Cells and in Lungs of Infected Mice

Stefanie Deinhardt-Emmer et al.

Summary: The PI3K inhibitor Pictilisib has been identified as a potent inhibitor of IV propagation, showing reduced viral titers and inflammatory response in the lungs, making it a promising candidate for anti-IV therapies. Repurposing previously approved substances is a cost-effective and efficient way for the development of novel antiviral strategies.
