Cocaine Triggers Astrocyte-Mediated Synaptogenesis

Summary: The study shows that the developmental synaptogenic mechanism mediated by astrocyte-released thrombospondins (TSPs) and their neuronal receptor alpha 2 delta-1 is utilized during cocaine experience to induce spinogenesis and the generation of AMPA receptor-silent glutamatergic synapses in the adult nucleus accumbens shell (NAcSh). Further research indicates that disruption of TSP2-alpha 2 delta-1-mediated spinogenesis and synapse generation in NAcSh reduces cue-induced cocaine seeking after withdrawal from cocaine self-administration and cue-induced reinstatement of cocaine seeking after drug extinction.

Heroin Seeking and Extinction From Seeking Activate Matrix Metalloproteinases at Synapses on Distinct Subpopulations of Accumbens Cells

Summary: This study found that heroin-associated cues transiently increased MMP-9 activity around D1-MSNs, while extinction training induced long-lasting increases in MMP-2 activity around D2-MSNs. Moreover, heroin cues also increased tissue inhibitor of metalloproteinases (TIMP-1,2) which temporarily inhibited MMP-2 activity around D2-MSNs during cue-induced heroin seeking. The differential regulation of heroin seeking and extinguished seeking by different MMP subtypes on distinct cell populations emphasizes the importance of MMP-2,9 activity in heroin-induced cell-specific synaptic plasticity.

Summary: Studies on the neurobiological underpinnings of substance use disorder (SUD) have revealed specific brain regions necessary for behaviors driven by drugs versus natural rewards. While many brain regions are involved in behaviors motivated by all types of rewards, certain interconnected regions are selectively necessary for drug-seeking behaviors. Further research is needed to clarify the distinct roles of these brain regions in drug- versus natural reward-associated behaviors.

Effects of Gabapentin on Dorsal Anterior Cingulate Cortex GABA and Glutamate Levels and Their Associations With Abstinence in Alcohol Use Disorder: A Randomized Clinical Trial

Summary: The study found that the effects of gabapentin on GABA and glutamate levels were significantly correlated with the participants' percent days abstinent during early treatment. Specifically, gabapentin was associated with greater increases in glutamate and greater decreases in GABA levels in participants who remained mostly or entirely abstinent early on, but with the opposite pattern in participants who drank on more than half of the days preceding the second scan.
