The roles of microRNAs in mouse development

Brian DeVeale et al.

Summary: MiRNAs play crucial regulatory roles in mouse development by negatively regulating multiple mRNA targets. They coordinate cell fate transitions during development and contribute to organismal fitness through homeostatic roles in adults. Recent advances in miRNA-knockout phenotypes and related targets, dosage, and interactions have enhanced our understanding of their roles in mammalian development and adaptive responses.

A pan-cancer atlas of somatic mutations in miRNA biogenesis genes

Paulina Galka-Marciniak et al.

Summary: The levels of many miRNAs are differentiated in cancer, and the study identified some miRNA biogenesis genes that are over-mutated in specific cancers and associated with patient survival. Hotspot mutations in DICER1 were found to affect the levels of miRNAs, ultimately creating an atlas of miRNA biogenesis alterations.

isomiRs-Hidden Soldiers in the miRNA Regulatory Army, and How to Find Them?

Ilias Glogovitis et al.

Summary: Research on microRNAs (miRNA) in cancer and other diseases has led to the development of diverse computational approaches and experimental methods for predicting and validating their biological and clinical significance as disease biomarkers. The use of next-generation deep sequencing has allowed for the discovery of new RNA biomarkers, including diverse sequence variants of mature miRNAs or isomiRs. However, challenges and pitfalls exist in the analysis of isomiR expression.

Small RNAs are modified with N-glycans and displayed on the surface of living cells

Ryan A. Flynn et al.

Summary: This study challenges the conventional view that RNA is not a major target of glycosylation by providing evidence that mammals use RNA as a third scaffold for glycosylation. The researchers found that conserved small noncoding RNAs bear sialylated glycans, which are present in multiple cell types and mammalian species. The majority of glycoRNAs were located on the cell surface and were shown to interact with anti-dsRNA antibodies and members of the Siglec receptor family, suggesting a direct interface between RNA biology and glycobiology.

Cell-type-specific profiling of loaded miRNAs from Caenorhabditis elegans reveals spatial and temporal flexibility in Argonaute loading

Christopher A. Brosnan et al.

Summary: Researchers have generated a genome-wide map of active miRNAs in Caenorhabditis elegans, revealing cell-type-specific patterns of miRNAs loaded into Argonaute silencing complexes. This study demonstrates spatial and temporal dynamicism, flexibility of miRNA loading, and suggests miRNA regulatory mechanisms via AGO selectivity in different tissues and during ageing. Additionally, widespread changes in AGO-regulated gene expression were resolved by analyzing translatomes specifically in neurons, providing insights into AGO loading flexibility.
