Serum citrullinated histone H3 concentrations differentiate patients with septic verses non-septic shock and correlate with disease severity

Yuzi Tian et al.

Summary: The study showed that levels of CitH3 are elevated in patients with septic shock and correlates with PAD2 and PAD4 concentrations, disease severity, and prognosis. Furthermore, higher serum concentrations of CitH3 at 24 hours and 48 hours were found in septic patients who did not survive, suggesting a potential role of CitH3 as a prognostic biomarker for septic shock.

Peptidylarginine Deiminase Inhibition Prevents Diabetes Development in NOD Mice

Fernanda M. C. Sodre et al.

Summary: In this study, systemic inhibition of peptidylarginine deiminases (PADs) through BB-Cl-amidine treatment prevented diabetes development in NOD mice by reducing citrullination levels in the pancreas, decreasing circulating autoantibody titers, and reducing neutrophil extracellular trap formation. Additionally, the treatment induced a shift in cytokine production and increased regulatory T cell frequency. This highlights the potential of PAD inhibition for treating autoimmune diseases like type 1 diabetes.

Citrullinated myelin induces microglial TNFα and inhibits endogenous repair in the cuprizone model of demyelination

Miranda M. Standiford et al.

Summary: This study revealed that citrullinated myelin induces a unique microglial response with increased production of tumor necrosis factor alpha (TNF alpha). Injection of citrullinated myelin inhibits spontaneous remyelination significantly, while neutralization of TNF alpha restores remyelination to normal levels.

Citrullination of Amyloid-β Peptides in Alzheimer's Disease

Soumya Mukherjee et al.

Summary: The study identified citrullination of A beta in sporadic and familial Alzheimer's disease brains using high-resolution mass spectrometry. Results demonstrated a common mechanism for citrullination of A beta in both types of AD. Citrullination of A beta is a remarkably common post-translational modification closely associated with A beta formation and accumulation in AD pathology.

Citrullination as a novel posttranslational modification of matrix metalloproteinases

Lise Boon et al.

Summary: Citrullination of MMPs by PADs is a novel posttranslational modification that may impact their activities and functions. Citrullinated MMP-9 exhibits increased affinity for gelatin and more efficient activation compared to control MMP-9. Furthermore, other MMPs such as MMP-1 can also be efficiently citrullinated by PADs.
