High-content imaging-based pooled CRISPR screens in mammalian cells

Xiaowei Yan et al.

Summary: A microscopy-based optical enrichment approach was developed for selecting and marking cells displaying specific CRISPR-induced phenotypes, followed by isolation using fluorescence-activated cell sorting (FAG). Automated phenotypic identification and photoactivation of cells were achieved through a plugin developed for the open source software mu Manager, allowing screening of approximately 1.5 million individual cells in 8 hours. This scalable approach facilitated the identification of 14 bona fide hits out of 6,092 sgRNAs targeting 544 genes in terms of their effects on nuclear size regulation.

Actin on and around the Nucleus

Patricia M. Davidson et al.

Summary: Actin plays crucial roles in various cellular processes, and recent research has uncovered the potential for actin to transmit force directly to the nucleus and genome. Ongoing studies are elucidating the mechanisms involved and revealing novel functions of actin in mitosis, meiosis, gene expression, and cellular responses to mechanical stimuli.

Rad52 SUMOylation functions as a molecular switch that determines a balance between the Rad51- and Rad59-dependent survivors

Ferose Charifi et al.

Summary: The study reveals that in yeast lacking telomerase, functional telomeres can be restored by rare recombination events, and demonstrates that SUMOylation plays a differential role in type I and type II survivors.
