The potential of diagnostic point-of-care tests (POCTs) for infectious and zoonotic animal diseases in developing countries: Technical, regulatory and sociocultural considerations

Emma C. Hobbs et al.

Summary: Remote and rural communities in low- and middle-income countries are disproportionately affected by infectious animal diseases due to their close contact with livestock and limited access to animal health personnel. Point-of-care tests (POCTs) have the potential to disrupt traditional animal health surveillance paradigms in these countries, but many current POCTs target only a small number of key agricultural and zoonotic animal diseases. Evaluation and validation of POCTs in LMICs are lacking, posing challenges for potential customers and potentially undermining their effectiveness in disease control. Further research and regulatory measures are urgently needed to fully realize the potential of animal disease POCTs in LMICs.

Development of a fluorescent probe-based real-time reverse transcription recombinase-aided amplification assay for the rapid detection of classical swine fever virus

Fei Tu et al.

Summary: A novel fluorescent probe-based rRT-RAA assay targeting a conserved region of CSFV was developed, showing high specificity and sensitivity for CSFV detection. The assay exhibited good reproducibility and comparable sensitivity to traditional RT-qPCR, with a high coincidence rate between the two assays. The amplification products could be directly observed with naked eyes under a portable blue light imager, indicating the potential for on-site testing.

An Isothermal Molecular Point of Care Testing for African Swine Fever Virus Using Recombinase-Aided Amplification and Lateral Flow Assay Without the Need to Extract Nucleic Acids in Blood

Yuhang Zhang et al.

Summary: In this study, a sensitive, specific, rapid, and simple molecular point of care testing for African swine fever virus (ASFV) B646L gene in blood samples was established without the need to extract viral DNA, providing a good choice for ASF diagnosis in the future. The method showed 100% coincident rate with OIE-recommended PCR, had higher sensitivity, and exhibited good specificity in clinical blood samples evaluation.
