Summary: The humoral immune response plays a key role in suppressing SARS-CoV-2, and the ability of antibodies to disrupt the binding of the viral spike protein to ACE2 receptor is controlled by antibody affinity. Utilizing microfluidic antibody-affinity profiling, the serum-antibody mediated inhibition of ACE2-spike binding can be quantified in SARS-CoV-2 seropositive individuals. These experiments introduce a new type of in-solution receptor binding competition assay with potential applications in convalescent plasma therapy, therapeutic antibody development, and vaccine development.
