DeepDILI: Deep Learning-Powered Drug-Induced Liver Injury Prediction Using Model-Level Representation

Ting Li et al.

Summary: The DeepDILI model, developed using drugs approved before 1997, outperformed conventional ML algorithms in predicting DILI potential of newly approved drugs. It showed significant performance improvement compared to molecule-based deep neural networks and identified 21 enriched chemical descriptors associated with DILI outcomes. The DeepDILI model based on Mold2 descriptors demonstrated higher discrimination power for drugs in the 'alimentary tract and metabolism' category and outperformed models with Mol2vec and MACCS descriptors.

DeepCellState: An autoencoder-based framework for predicting cell type specific transcriptional states induced by drug treatment

Ramzan Umarov et al.

Summary: DeepCellState is a deep learning framework that predicts cell type specific transcriptional states after drug treatment based on a large collection of transcriptional drug perturbation profiles. The method shows improved accuracy when generalized to multiple cell types, even for treatments with drugs not seen during training. Analysis of the trained model reveals the ability to learn regulatory relationships between genes in a fully data-driven manner.

Safer chemicals using less animals: kick-off of the European ONTOX project

Mathieu Vinken et al.

Summary: The 3Rs concept, focusing on replacement, reduction, and refinement of animal experimentation, is gaining attention worldwide and has been incorporated into legislation, particularly in the European Union. The ONTOX project in Europe aims to develop AI-driven methods for repeated dose toxicity testing of chemicals without the use of animals to advance human risk assessment. This project will have a significant impact on consolidating Europe's leading position in the development and application of animal-free methods for chemical risk assessment.

Leveraging high-throughput screening data, deep neural networks, and conditional generative adversarial networks to advance predictive toxicology

Adrian J. Green et al.

Summary: This study demonstrates the use of a combined deep neural network (DNN) and conditional Generative Adversarial Network (cGAN) to predict the toxicity of untested compounds by leveraging a large set of experimental toxicity data and chemical structure information, showing high accuracy and effectiveness.

The Tox21 10K Compound Library: Collaborative Chemistry Advancing Toxicology

Ann M. Richard et al.

Summary: The Tox21 project has conducted large-scale chemical screening since 2009, establishing the largest compound library to date, with specialized resources and capabilities from federal partners enriching the data. Different goals among partners led to three overlapping compound libraries, which complement each other and broaden the coverage range.
