GLORYx: Prediction of the Metabolites Resulting from Phase 1 and Phase 2 Biotransformations of Xenobiotics

Christina de Bruyn Kops et al.

Summary: Predicting metabolite structures in humans using GLORYx, which combines machine learning-based SoM prediction and reaction rule sets, is beneficial for drug development. GLORYx achieved a recall of 77% and an AUC of 0.79 on a test dataset, but found it more challenging to rank predicted metabolites for phase 2 than for phase 1 metabolism.

Identification and structure characterization of five synthetic opioids: 3,4-methylenedioxy-U-47700,o-methyl-acetylfentanyl, 2-thiophenefentanyl, benzoylfentanyl and benzoylbenzylfentanyl

Magdalena Poplawska et al.

Summary: The increasing number of new synthetic opioids, especially fentanyl analogs, pose a serious threat to public health due to their high potency, ease of addiction, and risk of overdose. This study successfully employed LC-QTOF-MS/MS and NMR techniques for the unambiguous identification of five synthetic opioids, providing important reference data for forensic and clinical laboratories.
