Exploring the Trans-Cleavage Activity of CRISPR/Cas12a on Gold Nanoparticles for Stable and Sensitive Biosensing

Xiaoyi Fu et al.

Summary: Taking advantage of the trans-cleavage activity of CRISPR/Cas12a on gold nanoparticles, a new spherical nucleic acid (SNA) reporter has been developed for stable and sensitive CRISPR-Dx biosensing. Different trans-cleavage activities of Cas12a systems were discovered on a nanoparticle surface, with LbCas12a highly dependent on DNA strand density and length. The SNA reporter exhibited improved stability and high sensitivity, showing potential for future research in biology science and medical diagnoses.

MXene Coupled with CRISPR-Cas12a for Analysis of Endotoxin and Bacteria

Anzhi Sheng et al.

Summary: In this study, it was found that MXene can efficiently adsorb single-stranded DNA and be coupled with CRISPR-Cas12a for sensitive detection of LPS and bacteria. Through carefully designed aptamers, the trans-cleavage activity of CRISPR-Cas12a is activated, allowing for selective and sensitive quantification of targets in different samples. This method provides a new direction for the exploration of universal analytical methods based on MXene coupled with CRISPR-Cas12a.

G-triplex: A new type of CRISPR-Cas12a reporter enabling highly sensitive nucleic acid detection

Tao Li et al.

Summary: By utilizing G-triplex (G3) as a reporter gene, G-CRISPR achieves a 20-fold improvement in analytical sensitivity, enabling detection of unamplified and amplified DNA as low as 50 pmol and 0.1 amol (one copy/reaction) respectively. The results demonstrate high specificity and sensitivity, and suggest that G-CRISPR could serve as a rapid, sensitive, and reliable biosensor in biomedical diagnostics.
