Summary: The study uncovers a hidden form of inhibitory synaptic plasticity that prevents the accumulation of excitatory long-term potentiation (LTP). Induction of excitatory LTP relocates α5-GABARs to inhibitory synapses, interrupting further LTP induction. This dual plasticity between inhibitory and excitatory synapses may be a critical early step in memory preservation.

Summary: The decision between long-term potentiation (LTP) and depression (LTD) in higher brain functions is determined by the complex cross-regulation of T286 and T305/306 autophosphorylation within the 12meric CaMKII holoenzyme. T305/306 phosphorylation selectively promotes LTD while T286 phosphorylation is required for both LTP and LTD. The differential T305/306 phosphorylation directs CaMKII movement to either excitatory or inhibitory synapses, coordinating plasticity at both synapse types.

Light-guided intrabodies for on-demand in situ target recognition in human cells

Summary: Fluorescently labeled nanobodies are ideal probes for visualizing intracellular targets at an endogenous level due to their high stability and specificity in living cells. A strategy to prevent premature binding through the development of photo-conditional intrabodies has been reported. These light-guided intrabodies offer high spatiotemporal precision in binding target proteins inside living cells, providing a versatile platform for downstream imaging and regulation of target proteins.
