The actin polymerization factor Diaphanous and the actin severing protein Flightless I collaborate to regulate sarcomere size

Su Deng et al.

Summary: The protein Dia plays a critical role in regulating sarcomere size by interacting with FliI, ensuring normal muscle function. These actin regulatory factors generate uniformly sized sarcomeres tuned for muscle contractions required for flight.

A light way for nuclear cell biologists

Giada Forlani et al.

Summary: The nucleus, a complex organelle in eukaryotic cells, plays a crucial role in safeguarding and managing genetic information. Recent discoveries of a dynamic nuclear cytoskeleton made of actin filaments have opened up new insights into processes like gene expression and DNA repair. Optogenetics, a state-of-the-art technology using genetically encoded light sensitive proteins, offers exciting possibilities for studying nuclear cell biology.

High-content imaging-based pooled CRISPR screens in mammalian cells

Xiaowei Yan et al.

Summary: A microscopy-based optical enrichment approach was developed for selecting and marking cells displaying specific CRISPR-induced phenotypes, followed by isolation using fluorescence-activated cell sorting (FAG). Automated phenotypic identification and photoactivation of cells were achieved through a plugin developed for the open source software mu Manager, allowing screening of approximately 1.5 million individual cells in 8 hours. This scalable approach facilitated the identification of 14 bona fide hits out of 6,092 sgRNAs targeting 544 genes in terms of their effects on nuclear size regulation.

Polo-like kinase 1 independently controls microtubule-nucleating capacity and size of the centrosome

Midori Ohta et al.

Summary: PLK1 not only controls the expansion of PCM, but also independently regulates the generation of binding sites for gamma-tubulin complexes on the PCM matrix. Inhibiting PLK1-dependent gamma-tubulin docking sites leads to spindle defects and impaired chromosome segregation, highlighting the importance of phospho-regulated centrosomal gamma-tubulin docking sites in spindle assembly.

Drosophila female germline stem cells undergo mitosis without nuclear breakdown

Tingting Duan et al.

Summary: This study demonstrates that compromised nuclear lamina integrity in Drosophila germ cells can activate checkpoint kinases, leading to loss of germline stem cells. The structural defects in nuclear lamina during mitosis are linked to checkpoint activation, affecting the normal function of germ cells.

Acto-myosin network geometry defines centrosome position

Ana Joaquina Jimenez et al.

Summary: The position of the centrosome within a cell is not always at the geometric center, but can be close to the cell edge. Centrosome positioning appears to respond accurately to the architecture and anisotropy of the actin network, rather than the shape of the cell.

Integrated spatial genomics reveals global architecture of single nuclei

Yodai Takei et al.

Summary: This study utilized DNA seqFISH+ technology to image 3,660 chromosomal loci in single mouse embryonic stem cells, along with observing 17 chromatin marks and subnuclear structures by sequential immunofluorescence and the expression profile of 70 RNAs. The results suggest that many loci are consistently associated with immunofluorescence marks and form "fixed points" in nuclear organizations in single cells, while highly expressed genes appear to be pre-positioned to active nuclear zones. The research also uncovered distinct mouse ES cell subpopulations with characteristic combinatorial chromatin states.

Muscle repair after physiological damage relies on nuclear migration for cellular reconstruction

William Roman et al.

Summary: The study revealed that localized muscle injuries triggered by exercise activate a myofiber self-repair mechanism that is independent of satellite cells. A signaling cascade involving calcium, Cdc42, and phosphokinase C attracts myonuclei to the damaged site, accelerating sarcomere repair and delivering messenger RNA for cellular reconstruction. Myofiber self-repair is a cell-autonomous protective mechanism, offering an alternative model for understanding muscle architecture restoration in health and disease.

Molecular mechanism of cytokinin-activated cell division in Arabidopsis

Weibing Yang et al.

Summary: Mitogens trigger cell division in animals, while in plants, cytokinins derived from adenine stimulate cell proliferation. Cytokinin signaling in Arabidopsis SAM is shown to regulate cell division by promoting nuclear shuttling of MYB3R4, a transcription factor activating mitotic gene expression. Rapid nuclear accumulation of MYB3R4 at G2-to-M transition, associated with a peak in cytokinin concentration, initiates a transcriptional cascade driving mitosis and cytokinesis, mimicking the effects of enhanced cell proliferation and meristem growth.

Regulation of apical constriction via microtubule-and Rab11-dependent apical transport during tissue invagination

Thao Phuong Le et al.

Summary: The study reveals that MT-dependent intracellular trafficking regulates apical constriction during salivary gland invagination in Drosophila embryos. Key components involved in protein trafficking, such as Rab11 and Nuclear fallout, are crucial for apicomedial myosin formation and apical constriction. MTs and Rab11 are essential for the apical enrichment of key proteins like Fog ligand, Crumbs, and E-Cadherin, which play a role in coordinating cell behaviors during tubular organ formation.

Asymmetric nuclear division in neural stem cells generates sibling nuclei that differ in size, envelope composition, and chromatin organization

Chantal Roubinet et al.

Summary: Studying the process in Drosophila neural stem cells, researchers find that the nuclear compartment boundary is maintained during mitosis by a supporting nuclear lamina. This mitotic nuclear envelope is then asymmetrically remodeled and partitioned to give rise to daughter nuclei with different envelope compositions and sizes, which is influenced by asymmetric nuclear growth and resealing processes.

Linker histone H1.8 inhibits chromatin binding of condensins and DNA topoisomerase II to tune chromosome length and individualization

Pavan Choppakatla et al.

Summary: In this study, the role of linker histone H1.8 in regulating mitotic chromosome organization is highlighted. H1.8 antagonizes condensins and topo II, affecting loop extrusion and chromosome individualization, ultimately influencing chromosome morphology and organization. The findings suggest that H1.8 acts as a key regulator in maintaining chromosome integrity during mitosis by modulating the functions of condensins and topo II.
