Summary: In this study, the chromosome-scale reference genome of Astragalus sinicus (CMV) was generated, which provided valuable insights into the genetic basis of nodulation and symbiosis in CMV. It was found that CMV has undergone two whole-genome duplication events, which shaped the genes involved in rhizobial symbiosis and hormonal regulation of nodulation. Additionally, resistance genes were found to be more highly expressed in roots than in nodules, indicating a potential role in enhancing plant immunity in roots against pathogen infection.
