Summary: Cas12a is a versatile nuclease enzyme in the CRISPR-Cas system, composed of REC and Nuc lobes with the BH domain playing a critical role in its enzymatic activity and conformational transitions. BH variants have been shown to impact the trimming activity, cleavage accuracy and conformational preferences of Cas12a, favoring the open state. Additionally, insights into the role of BH in the structural dynamics of FnCas12a have been uncovered through biochemical and single-molecule FRET studies.
