The Portal Vertex of KSHV Promotes Docking of Capsids at the Nuclear Pores

Daniela Dunn-Kittenplon et al.

Summary: This study investigates the docking of KSHV capsids at NPCs, finding a significantly lower attachment rate of ORF43-null capsids to the nuclear envelope compared to wild-type capsids. The study concludes that although not essential, the portal plays a role in mediating the interaction of CVSC proteins with Nups, suggesting a model where WT capsids can dock at the nuclear envelope through a non-portal penton vertex, leading to a potential infection 'dead end'.

UL25 Capsid Binding Facilitates Mechanical Maturation of the Herpesvirus Capsid and Allows Retention of Pressurized DNA

Krista G. Freeman et al.

Summary: Viruses undergo a maturation process to meet conflicting virion assembly requirements. Herpesviruses achieve capsid reinforcement and stable DNA encapsidation through a mechanical capsid maturation process facilitated by multifunctional auxiliary protein UL25. Gradual capsid reinforcement by progressive UL25 binding is a feasible mechanism correlated with DNA packaging progression, presenting a new antiviral strategy resistant to drug resistance.

Navigating the Cytoplasm: Delivery of the Alphaherpesvirus Genome to the Nucleus

Gregory A. Smith

Summary: Herpesviruses, including alphaherpesviruses, deliver their genetic content to nuclei upon entering cells. The viral envelope fuses with a cell membrane, depositing the capsid and tegument proteins into the cytosol. While the capsid is not the effector of genome delivery, tegument proteins play a crucial role in guiding it to the nucleus, increasing cell susceptibility to infection. Cells in mucosal epithelium and neurons provide a favorable niche for alphaherpesviruses, which have adapted to efficiently invade the nervous system and establish infection.
