Mechanism of Lamellar Body Formation by Lung Surfactant Protein B

Summary: This study reveals the importance of different domains of the SP-B protein in the formation of lamellar bodies (LBs) in the lungs. The N-terminal domain plays a key role in the export of pre-proSP-B from the endoplasmic reticulum (ER) and its sorting to LBs, while the C-terminal domain facilitates ER export. The middle domain, generated after proteolytic cleavage, is responsible for the formation of the concentric membrane layers characteristic of LBs.

Heterogeneity in Human Induced Pluripotent Stem Cell-derived Alveolar Epithelial Type II Cells Revealed with ABCA3/SFTPC Reporters

Summary: In this study, a new ABCA3:GFP reporter was generated for the purification of induced pluripotent stem cell-derived alveolar epithelial type 2 cells (iAEC2s) and visualization of organelles associated with AEC2 maturation. Comparative RNA sequencing and functional studies revealed cellular heterogeneity within the iAEC2 population, with ABCA3:GFP single-positive cells exhibiting faster growth kinetics and higher pluripotency compared to ABCA3:GFP/SFTPCtdTomato double-positive cells, which expressed more maturation markers.
