HPS6 Regulates the Biogenesis of Weibel-Palade Body in Endothelial Cells Through Trafficking v-ATPase to Its Limiting Membrane

Jiran Lu et al.

Summary: This study discovered that HPS6 is involved in the biogenesis of WPBs by transporting v-ATPase to the WPB membrane, maintaining its acidic luminal pH critical for vWF tubule formation.

Genetic variants and mutational spectrum of Chinese Hermansky-Pudlak syndrome patients

Teng Liu et al.

Summary: Hermansky-Pudlak syndrome (HPS) is a rare recessive disorder characterized by various symptoms, with HPS-1 being the most common subtype. 11 causative genes have been identified in humans, with the first case of HPS-2 found in the Chinese population. The study results will help evaluate the relationship between HPS genotypes and phenotypes.

Inflammatory bowel disease in Hermansky-Pudlak syndrome: a retrospective single-centre cohort study

Summary: This study aimed to expand the understanding of IBD in patients with Hermsnky-Pudlak syndrome (HPS) by retrospectively analyzing records and clinical features of patients evaluated at the NIH Clinical Center. Results showed that some patients with HPS develop IBD resembling Crohn's disease, with genetic heterogeneity being a feature of HPS IBD. The study also identified HPS3 as a new gene associated with human IBD, and early-onset IBD can occur in patients with HPS.

A zinc transporter, transmembrane protein 163, is critical for the biogenesis of platelet dense granules

Yefeng Yuan et al.

Summary: Lysosome-related organelles (LROs) are crucial for ion homeostasis in cells, and defects in these organelles lead to platelet dense granules (DGs) deficiency in Hermansky-Pudlak syndrome (HPS), where a newly identified zinc transporter, TMEM163, plays a critical role.

Summary: Hermansky-Pudlak syndrome 9 (HPS-9) is a rare recessive disorder caused by BLOC1S6 gene mutations. This case report of the first HPS-9 patient in a Chinese population identified novel variants in the BLOC1S6 gene, expanding the mutational spectrum of HPS genes.

A BLOC-1-AP-3 super-complex sorts a cis-SNARE complex into endosome-derived tubular transport carriers

Shanna L. Bowman et al.

Summary: The vSNARE VAMP7 is sorted into transport carriers in complex with the tSNARE STX13, with sorting requiring recognition by either AP-3 delta or pallidin subunit of BLOC-1, but not both. This mechanism allows for redundant recognition of sorting determinants on a SNARE complex by an AP-3-BLOC-1 super-complex.

AP-3-dependent targeting of flippase ATP8A1 to lamellar bodies suppresses activation of YAP in alveolar epithelial type 2 cells

Seunghyi Kook et al.

Summary: Lamellar bodies (LBs) are lysosome-related organelles (LROs) in AT2 cells that play a crucial role in cell homeostasis. Disruption of trafficking pathways mediated by AP-3 can lead to pulmonary fibrosis through affecting the transport of ATP8A1 to LBs.

Melanosome Biogenesis in the Pigmentation of Mammalian Skin

Linh Le et al.

Summary: Melanins, the main pigments of the skin and hair in mammals, are synthesized within membrane-bound organelles of melanocytes called melanosomes. The structure and function of melanosomes are determined by resident transmembrane proteins, defects of which may lead to various forms of albinism. Further investigation is needed to address unanswered questions regarding the molecular mechanisms of melanosome formation and the delivery of specific components.
