The spinosyns, spinosad, spinetoram, and synthetic spinosyn mimics - discovery, exploration, and evolution of a natural product chemistry and the impact of computational tools

Thomas C. Sparks et al.

Summary: Spinosyns, derived from soil inhibiting microorganisms, have been developed into various insect control products over the years with the help of artificial intelligence and computational tools. The recent synthetic spinosyn mimics have shown promising insecticidal effects against lepidopteran pests.

The cys-loop ligand-gated ion channel gene superfamilies of the cockroaches Blattella germanica and Periplaneta americana

Andrew K. Jones et al.

Summary: The cysLGIC superfamilies of Blattella germanica and Periplaneta americana are the largest insect cysLGIC superfamilies characterized to date, with 30 and 32 subunit-encoding genes, respectively. These cockroaches possess ion channels predicted to be gated by acetylcholine, gamma-aminobutyric acid, glutamate, histamine, as well as orthologues of the drosophila pH-sensitive chloride channel (pHCl), CG8916 and CG12344. The diversity of the cockroach cysLGICs is broadened by alternative splicing and RNA A-to-I editing.

Insecticide resistance management and industry: the origins and evolution of the Insecticide Resistance Action Committee (IRAC) and the mode of action classification scheme

Thomas C. Sparks et al.

Summary: IRAC, established in 1984, aims to address insecticide resistance issues in the crop protection industry. It has grown from a few agrochemical companies to a global group providing informational and educational tools for resistance management.

Role of nicotinic acetylcholine receptor subunits in the mode of action of neonicotinoid, sulfoximine and spinosyn insecticides in Drosophila melanogaster

Trent Perry et al.

Summary: This study systematically investigates the insect response to different classes of insecticides and highlights the specificity of receptor subunits in insecticide resistance. The findings provide new insights into the mode of action of insecticides and reveal a strong conservation of subunit targets across different insect species.

Mutation (G275E) of nAChR subunit Foα6 associated with spinetoram resistance in Australian western flower thrips, Frankliniella occidentalis (Pergande)

Yizhou Chen et al.

Summary: Research identified resistance to spinetoram in Australian Western flower thrips due to a G275E mutation in the nicotinic acetylcholine receptor gene. The unexpectedly wide distribution of the resistance allele may be linked to extensive chemical use in nearby crops. The development of a molecular diagnostic assay provides timely and accurate resistance frequency information to support sustainable chemical use.

Agonist and antagonist properties of an insect GABA-gated chloride channel (RDL) are influenced by heterologous expression conditions

Charles L. C. Smelt et al.

Summary: This study cloned the analogous gene BtRDL from bumblebees and examined its pharmacological properties, finding that its sensitivity to GABA was influenced by heterologous expression conditions. The phenomenon was not observed with DmRDL cloned from fruit flies, and changes in apparent affinity were also observed with co-expression of chaperone protein NACHO. Additionally, expression-dependent changes in apparent affinity were observed with both an allosteric agonist and a non-competitive antagonist, indicating a broader impact on pLGICs.
