Regulatory T-cell phenotypes in children with sickle cell disease

Summary: Among children with SCD, HU treatment exhibited significant qualitative and quantitative effects on Tregs. HU treatment in SCD decreases the frequency of Tregs, as well as the levels of the most suppressive Tregs: HLA-DR+, CD39+, and CD69+. At the same time, HU increases the proportion of naive CD45RA+ Tregs. Our study showed the impact of HU therapy on Tregs in children with SCD.

Summary: Sickle cell disease serves as a prime example of bidirectional translational research, with decades of bench research translating into new drugs and genetic therapies based on clinical observations of abnormal red blood cells. The introduction of hydroxyurea therapy and research on its fetal hemoglobin induction mechanism have paved the way for new HbF-reactivating agents. Understanding the molecular mechanisms behind HU's effects has also contributed to the development of recently approved anti-sickling drugs, reflecting a truly bidirectional translation from bench to bedside.
