SCSIO 03219, involved a series of transformations from epichlorohydrin to an ent-estrane, then to a nat-androstane, ultimately establishing the natural product target. The success of these studies relied on the implementation of two rearrangement processes to formally invert the quaternary center at C13 and establish the C10 quaternary center.

Summary: This highly enantioselective vinylogous Mukaiyama aldol reaction to ketoesters is catalyzed by a hydrogen-bond-donor-based bifunctional organocatalyst. Addition of silyloxy dienol ether yields multifunctional chiral tertiary alcohols with a versatile α, β-unsaturated aldehyde and excellent enantiocontrol.

Summary: A catalytic asymmetric sequential annulation/aldol reaction of 4-isothiocyanato pyrazolones and allenyl ketones has been developed, leading to a series of spiro[pyrrole-pyrazolone] heterocycles and structurally novel spiro[thiopyranopyrrole-pyrazolone] derivatives in good yields with high to excellent enantioselectivities. The diversity of product structures was further enhanced by easy transformations.
