Upregulation of histone acetylation reverses organic anion transporter 2 repression and enhances 5-fluorouracil sensitivity in hepatocellular carcinoma

Yingying Wang et al.

Summary: Hepatocellular carcinoma (HCC) is a common malignant tumor globally, with chemotherapy resistance often attributed to abnormal drug transporters. The study identified histone hypoacetylation as the mechanism behind OAT2 downregulation in HCC, and demonstrated that histone deacetylases inhibitor can reverse this state, enhancing drug uptake and sensitivity to chemotherapy in HCC cells.

Identification of the key target profiles underlying the drugs of narrow therapeutic index for treating cancer and cardiovascular disease

Jiayi Yin et al.

Summary: This study analyzed the target property of NTI drugs in cancers and cardiovascular disorders by calculating biological system profiles and human protein-protein interaction network properties. Differentiated features between the two diseases were discovered using an AI-based algorithm, revealing distinct underlying mechanisms of NTI drugs. Ten shared features and four unique features were identified for both diseases to distinguish NTI from NNTI drug targets. These findings provide novel guidance in drug discovery and clinical research, helping to estimate the drug safety of cancer and cardiovascular disease.
