Multi-omics analysis suggests enhanced epileptogenesis in theCornu Ammonis3 of the pilocarpine model of mesial temporal lobe epilepsy

Amanda M. Canto et al.

Summary: Research using a multi-omics approach in an animal model of mesial temporal lobe epilepsy revealed differences in the transcriptomic and proteomic profiles between the dentate gyrus (DG) and Cornu Ammonis 3 (CA3) of the hippocampus, suggesting enhanced epileptogenesis in the CA3 region and abnormalities mainly occurring in CA3. Abnormal regulation of excitatory neuronal mechanisms mediated by NMDA receptors, changes in serotonin signaling, and neuronal activity controlled by CaMK regulation and LRRK2/WNT signaling pathways are involved in the epileptogenesis in the pilocarpine model.

NLRP3 and NLRP1 inflammasomes are up-regulated in patients with mesial temporal lobe epilepsy and may contribute to overexpression of caspase-1 and IL-β in sclerotic hippocampi

Eliana Cristina de Brito Toscano et al.

Summary: In patients with mesial temporal lobe epilepsy, up-regulation of NLRP1 and NLRP3 in sclerotic hippocampi is associated with elevated levels of IL-1 beta and caspase-1, while increased levels of peripheral caspase-1 are related to bilateral hippocampal sclerosis. The findings suggest a role for inflammasome activation in both central and peripheral inflammation in TLE.

Summary: This Review provides an overview of the evidence for daily, multi-day, and yearly cycles in epileptic brain activity, discussing the potential clinical applications and highlighting technological advances. The authors address the question of why seizures occur when they occur, synthesizing historical, modern, and animal studies to uncover the mechanistic underpinnings of these cycles.

Sublayer- and cell-type-specific neurodegenerative transcriptional trajectories in hippocampal sclerosis

Elena Cid et al.

Summary: Hippocampal sclerosis, a major neuropathological feature of temporal lobe epilepsy, is characterized by overactive superficial CA1 pyramidal neurons in epileptic rodents. Single-cell gene expression analysis reveals sublayer-specific transcriptomic signatures and robust microglial pro-inflammatory responses. Neurodegenerative signatures primarily involve superficial cells.
