Alzheimer's disease pathology: pathways between central norepinephrine activity, memory, and neuropsychiatric symptoms

Heidi I. L. Jacobs et al.

Summary: The locus coeruleus-norepinephrine system plays a pivotal role in understanding the links between AD pathology and cognitive deficits. Neuropsychiatric symptoms are strongly linked to norepinephrine metabolism and phosphorylated tau, while memory deficits are associated with norepinephrine via a combination of tau and inflammation-driven amyloidosis.

Determining Amyloid-beta Positivity Using F-18-AZD4694 PET Imaging

Joseph Therriault et al.

Summary: This study assessed multiple methods for determining an optimal cutoff for F-18-AZD4694 PET positivity, with good convergence among different approaches. The findings suggest that a threshold of 1.55 SUVR may have reliable discriminative accuracy for amyloid-beta positivity on PET imaging.

Association between neuromelanin-sensitive MRI signal and psychomotor slowing in late-life depression

Kenneth Wengler et al.

Summary: This study showed that dopamine function abnormalities are related to slowed gait speed in older adults with late-life depression. While NM-MRI signal could predict treatment response to L-DOPA, it did not demonstrate associations with baseline gait speed, processing speed, or depression severity. Early findings suggest an increase in the NM-MRI signal after 3 weeks of L-DOPA treatment, indicating potential as a marker for dopamine-related psychomotor slowing.

MRI-assessed locus coeruleus integrity is heritable and associated with multiple cognitive domains, mild cognitive impairment, and daytime dysfunction

Jeremy A. Elman et al.

Summary: The study found that reduced LC integrity is associated with widespread differences across cognitive domains, daytime sleep-related dysfunction, and increased risk for aMCI. These findings highlight the potential of MRI-based measures of LC integrity in early identification of AD risk in late-middle-aged adults.

In vivo and neuropathology data support locus coeruleus integrity as indicator of Alzheimer's disease pathology and cognitive decline

Heidi I. L. Jacobs et al.

Summary: Various autopsy studies have shown that the locus coeruleus (LC) is the initial site of hyperphosphorylated TAU aggregation, and as TAU accumulates in LC neurons, pathology spreads throughout the cortex. Lower LC integrity is associated with increased TAU deposition in the entorhinal cortex in older individuals, and LC pigmentation ratings correlate with LC neuronal density, particularly in advanced Braak stages. These findings suggest that LC integrity may be a promising indicator of early Alzheimer's disease processes and cognitive changes.
