Cellular complexity in brain organoids: Current progress and unsolved issues

Abed AlFatah Mansour et al.

Summary: Brain organoids, derived from pluripotent stem cells through self-organization, are three-dimensional neural aggregates that mimic certain brain regions. They present an exciting area of research for studying human brain development, function, and dysfunction in new ways. Strategies including complementing neuroectodermal models with non-neuronal cell types, as well as transplantation approaches for higher cellular complexity and long-term survival, have been discussed in recent discoveries.

Cross-Comparison of Human iPSC Motor Neuron Models of Familial and Sporadic ALS Reveals Early and Convergent Transcriptomic Disease Signatures

Ritchie Ho et al.

Summary: This study utilized single-cell RNA sequencing to analyze neural cells from ALS patients, addressing limitations caused by genetic and experimental variability. They identified unified and reproducible ALS signatures, classified cells anatomically, and discovered predictive ALS markers by relaxing statistical thresholds. This approach revealed early, convergent, and MN-resolved signatures of ALS.

Genetic analysis of amyotrophic lateral sclerosis identifies contributing pathways and cell types

Sara Saez-Atienzar et al.

Summary: Despite progress in genetic research on ALS, the molecular mechanisms of the disease remain not fully understood. A study involving genome-wide data and polygenic risk score approach identified key biological pathways and cell types related to ALS, revealing themes like neuron projection morphogenesis and signal transduction. The findings suggest that genetic risk in ALS is consistently linked to certain types of neurons and oligodendrocytes, and certain genes may play a role in the disease. The study concludes that diverse genetic causes of ALS converge on a smaller number of common pathways and cell types.

Neurodevelopmental defects and neurodegenerative phenotypes in human brain organoids carrying Parkinson's disease-linked DNAJC6 mutations

Noviana Wulansari et al.

Summary: Loss-of-function mutations of DNAJC6 have been found to cause key pathologic features of Parkinson's disease in human midbrain-like organoids, including mDA neuron degeneration, alpha-synuclein aggregation, increased neuronal firing frequency, and mitochondrial and lysosomal dysfunctions. The defects in DNAJC6-mediated endocytosis impair the WNT-LMX1A signal during mDA neuron development, leading to the generation of vulnerable neurons with pathologic manifestations. These findings provide insights into disease mechanisms and potential therapeutic interventions for DNAJC6-PD.
