Structural basis for non-radical catalysis by TsrM, a radical SAM methylase

Hayley L. Knox et al.

Summary: The crystal structures of a cobalamin-dependent radical S-adenosylmethionine (SAM) methylase reveal an unexpected mechanism involving substrate-assisted catalysis with the carboxylate group of SAM serving as a general base. Tryptophan 2C methyltransferase (TsrM) methylates C2 of the indole ring of l-tryptophan during biosynthesis of thiostrepton's quinaldic acid moiety. The structures of TsrM from Kitasatospora setae show an essential arginine residue near the cobalamin cofactor, and a [4Fe-4S] cluster ligated by a glutamyl residue and three cysteines in a canonical CXXXCXXC RS motif. Substrate-assisted catalysis is suggested by structures in the presence of substrates, where the carboxylate group of SAM acts as a general base to deprotonate N1 of the tryptophan substrate, facilitating the formation of a C2 carbanion.

Biosynthesis of Oxetanocin-A Includes a B12-Dependent Radical SAM Enzyme That Can Catalyze both Oxidative Ring Contraction and the Demethylation of SAM

Aoshu Zhong et al.

Summary: Oxetanocin-A biosynthesis relies on the cooperative action of enzymes OxsB and OxsA, producing an unknown byproduct during the reaction. The study indicates that OxsB enzyme may possess methyltransferase activity, shedding light on its potential function beyond the described biosynthetic pathway.

Summary: The study determined the stereochemical course of the methyl group during the biosynthesis of the antibiotic fosfomycin, showing retention of configuration in the overall reaction. This outcome supports a double-inversion process and provides a method for investigating the mechanisms of other novel enzymes.

PROXIMA-1 beamline for macromolecular crystallography measurements at Synchrotron SOLEIL

Leonard M. G. Chavas et al.

Summary: The PROXIMA-1 undulator beamline at Synchrotron SOLEIL is dedicated to biomolecular crystallography experiments and has a layout designed to favor anomalous data recording and studies of crystals with large cell dimensions. Over 12 years, it has accommodated a large number of shifts and visitors, resulting in numerous published scientific papers and coordinates deposited in the Protein Data Bank. The beamline is equipped with advanced beam-shaping equipment, a three-axis goniometer, an X-ray detector, and a sample exchange robot, allowing for standard experimental schemes as well as remote or `mail-in' access.

Radical SAM Enzymes and Ribosomally-Synthesized and Post-translationally Modified Peptides: A Growing Importance in the Microbiomes

Alhosna Benjdia et al.

Summary: In order to address the antibiotic resistance crisis, novel strategies are urgently needed. Natural products have played a significant role in the discovery of new antibiotics, with ribosomally synthesized and posttranslationally modified peptides (RiPPs) being considered promising drug scaffolds. Recent studies have highlighted the important role of RiPPs in the human microbiota.
