A multimodal cell census and atlas of the mammalian primary motor cortex

Edward M. Callaway et al.

Summary: This study presents a multimodal cell census and atlas of the mammalian primary motor cortex, integrating various molecular and spatial information to reveal a unified genetic landscape of cortical cell types. The results establish a mechanistic framework of neuronal cell-type organization by linking molecular genetic information with phenotypic properties.

Genetic dissection of the glutamatergic neuron system in cerebral cortex

Katherine S. Matho et al.

Summary: The study established genetic strategies and tools to dissect and fate-map subpopulations of pyramidal neurons based on their developmental and molecular programs. Through combinatorial strategies, subsets of pyramidal neurons defined by developmental origin, marker expression, anatomical location, and projection targets can be targeted for viral access. These strategies provide an experimental framework for understanding the hierarchical organization and developmental trajectory of subpopulations of pyramidal neurons.

Spatially resolved cell atlas of the mouse primary motor cortex by MERFISH

Meng Zhang et al.

Summary: A single-cell transcriptome-imaging method, MERFISH, was used to create a molecularly defined and spatially resolved cell atlas of the mouse primary motor cortex. Approximately 300,000 cells were profiled, revealing 95 neuronal and non-neuronal cell clusters and a complex spatial map. Integration of MERFISH with retrograde labelling showed that cortical projections from neurons formed a complex network with individual clusters projecting to multiple target regions.

Epigenomic diversity of cortical projection neurons in the mouse brain

Zhuzhu Zhang et al.

Summary: By combining retrograde labeling with single-nucleus DNA methylation sequencing, researchers were able to identify unique epigenetic signatures of projection neurons that corresponded to their laminar and regional location and projection patterns. These findings helped to distinguish intra-telencephalic cells projecting to different cortical targets and confirmed the correlation between L5ET cortical neurons and their axonal projections.

A taxonomy of transcriptomic cell types across the isocortex and hippocampal formation

Zizhen Yao et al.

Summary: This study provides a comprehensive analysis of cell types in the mammalian isocortex and hippocampal formation, revealing a shared neural circuit organization between the two structures. The research also uncovers large-scale variations in cell types along different dimensions within these brain regions.

Phenotypic variation of transcriptomic cell types in mouse motor cortex

Federico Scala et al.

Summary: The study combines single-cell transcriptomic, morphological, and electrophysiological characteristics to classify over 1,300 neurons from the mouse motor cortex, revealing extreme diversity in gene expression, morphology, and electrophysiology. While broad families of transcriptomic types exhibit distinct morpho-electric phenotypes, individual transcriptomic types within the same family show a continuum of variability in morphology and electrophysiology without clear boundaries between them, indicating that neuronal types in the neocortex do not always form discrete entities.
