Atomic structures of respiratory complex III2, complex IV, and supercomplex III2-IV from vascular plants

Summary: This study determined the atomic structures of mitochondrial complex III (CIII2), complex IV (CIV), and their supercomplex in Vigna radiata using single-particle cryoEM. The findings showed specific differences in plant complexes and challenged long-standing assumptions, providing new mechanistic hypotheses for proton translocation and complex interactions. The structures revealed coordinated movements and conformational heterogeneity within the complexes, offering insights into the unique features of plant respiration machinery.

Summary: Energy conversion in aerobic organisms involves electron transfer from low-potential donors to dioxygen through the membrane-bound respiratory chain. The formation of supercomplex CIII2CIV1/2 facilitates electron transfer by two-dimensional diffusion of cyt. c, increasing QH(2):O-2 oxidoreductase activity and suggesting a regulatory role for supercomplex formation in the respiratory chain.

Cryo-EM grid optimization for membrane proteins

Summary: Our study systematically investigated the effects of buffer components, blotting conditions, and grid types on membrane protein grid preparation. We found that aggregation was a common issue, which could be addressed by adjusting detergents, salt concentration, or reconstitution methods. Furthermore, we discussed strategies for achieving optimal ice thickness, particle coverage, and orientation distribution on both free ice and support films. Our findings provide a clear roadmap for comprehensive screening of conditions for cryo-EM grid preparation of membrane proteins.
