Unraveling the In Vivo Protein Corona

Johanna Simon et al.

Summary: The study highlights the different interactions between nanoparticles and blood proteins under in vivo and in vitro conditions, emphasizing the importance of in vivo protein corona studies. Additionally, the protein composition of the in vivo corona remains relatively stable over time. To mimic the in vivo scenario, an ex vivo approach using fresh whole blood from an animal was established.

Unfolded Protein Corona Surrounding Nanotubes Influence the Innate and Adaptive Immune System

Jun-Young Park et al.

Summary: The protein corona surrounding nanoparticles significantly influences the subsequent immune response, with highly unfolded protein corona structures correlated with activation of proinflammatory cytokines and innate immune responses. This study clarified that the immune response mediated by the protein corona is greatly dependent on the type of plasma proteins surrounding the nanoparticles.

How to effectively prepare a sample for bottom-up proteomic analysis of nanoparticle protein corona? A critical review

Joanna Kruszewska et al.

Summary: With the growing interest in the biomedical applications of inorganic nanoparticles (NPs) in recent decades, there is a need for detailed characterization of bio-nano interactions, particularly the protein corona formed on NP surfaces. Analytical methods capable of identifying corona proteins are essential, and bottom-up proteomics is commonly used for this purpose. The experiment's sample preparation, including separation techniques and digestion methods, plays a crucial role in the identification process, with distinct protocols leading to different outcomes. Standardization of protein corona identification protocols is needed to prevent preferential detection of certain proteins with specific digestion procedures.

The protein corona and its effects on nanoparticle-based drug delivery systems

Hanmei Li et al.

Summary: The formation of a protein corona (PC) on nanoparticles (NPs) in the blood can influence their behavior. Strategies to reduce PC formation or exploit it to enhance NP functions are discussed in this review. Studying protein-NP interactions can provide insights into their clinical activity in health and disease.

Analysis of Nanotoxicity with Integrated Omics and Mechanobiology

Tae Hwan Shin et al.

Summary: Nanoparticles in biomedical applications offer advantages due to their small size, but evaluating their potential adverse effects on health is necessary and challenging. Omics approaches, in combination with machine learning techniques, provide a more comprehensive and sensitive way to assess nanotoxicity. Additionally, mechanobiology methods can be applied to evaluate the biophysical changes induced by nanoparticles, enhancing the overall understanding of their safety implications.

Summary: A novel antigen delivery system based on polysaccharide-coated gold nanoparticles has been developed targeting antigen-presenting cells expressing Dectin-1. The system elicits strong in vivo immune responses through production of high-titer antibodies and activation of antigen-recognizing T-cells, showing favorable antitumor cytokine balance.

In vitro coronal protein signatures and biological impact of silver nanoparticles synthesized with different natural polymers as capping agents

Priyanka Srivastava et al.

Summary: Biopolymer-capped nanosilver particles were synthesized to study their haemocompatibility in vitro and in vivo. Different biopolymer-capped particles showed varying levels of aggregation and stability in biologically relevant media. In vitro cytotoxicity studies revealed size and concentration-dependent haemolytic activity, while in vivo studies showed minimal haemolysis effects and accumulation of particles in organs.

Can the biomolecular corona induce an allergic reaction?-A proof-of-concept study

Anne Muehe et al.

Summary: The study aimed to investigate whether patients with or without hypersensitivity reactions to ferumoxytol nanoparticles develop specific protein corona profiles. The results suggest that analyzing the biomolecular corona ex vivo may provide predictive information on severe allergic reactions to ferumoxytol nanoparticles, potentially allowing for the diagnosis of patients predisposed to an allergic reaction based on proteomic patterns in the corona.
