Newly synthesized (R)-carvone-derived 1,2,3-triazoles: structural, mechanistic, cytotoxic and molecular docking studies

Summary: In this study, natural (R)-carvone was efficiently used to synthesize several 1,2,3-triazole derivatives with good yields. The cytotoxic activity of the newly synthesized compounds was analyzed, showing medium antitumor activity against selected human cancer cells. Density Functional Theory and molecular docking were employed to determine the electronic properties and stability of the synthesized compounds during inhibition of the anti-apoptotic protein for killing cancer cells, confirming experimental results.

Design, synthesis, biological evaluation and molecular docking of new 1,3,4-oxadiazole homonucleosides and their double-headed analogs as antitumor agents

Summary: A novel series of homonucleosides and their double-headed analogs were designed and synthesized, characterized by various spectroscopic techniques, and screened for cytotoxic activities against four cell lines. The double-headed analogue 18 showed significant growth inhibition in all cell lines tested, inducing apoptosis through caspase-3/7 activation and cell-cycle arrest. Molecular docking studies confirmed that compound 18 activated caspase-3 via hydrogen bonding and hydrophobic interactions.

Thiazolidinone-linked1,2,3-triazoles with monoterpenic skeleton as new potential anticancer agents: Design, synthesis and molecular docking studies

Summary: A novel series of 1,2,3-triazole-thiazolidinone-carvone hybrid compounds were designed and synthesized, with moderate anti-proliferative activity against human cancer cell lines. The most active compounds were found to induce apoptosis through caspase-3/7 activation, with different cell cycle effects. Molecular docking confirmed the strong bonding between these compounds and caspace-3.

Summary: A series of diverse triazoles linked to the hydroxyl group of totarol were synthesized and evaluated for their anticancer activity, showing moderate activities against cancer cell lines. The study provides important insights for further development of anticancer drugs. Compound 3g exhibited significant cytotoxicity against specific cancer cell lines and activated caspase-3 while inhibiting tubulin.

The theoretical studies on molecular properties were in agreement with the experimental results.

Discovery of novel furo[2,3-d]pyrimidin-2-one-1,3,4-oxadiazole hybrid derivatives as dual antiviral and anticancer agents that induce apoptosis

Summary: A new series of furo[2,3-d]pyrimidine-1,3,4-oxadiazole hybrids were synthesized using a multi-step eco-friendly approach and a one-pot Songoashira-heterocyclization protocol with nanostructured palladium pyrophosphate as a catalyst. These hybrids demonstrated broad-spectrum activity against varicella-zoster virus, with compound 9b showing higher potency and selectivity. Additionally, the hybrids exhibited moderate cytotoxicity in human cancer cell lines and induced apoptosis through caspase activation and cell cycle arrest.
