Mass spectrometry-based characterization of histones in clinical samples: applications, progresses, and challenges

Summary: In the past 15 years, research has shown the potential of investigating histone post-translational modifications and variants in clinical samples for discovering biomarkers and therapeutic targets in cancer biology. Mass spectrometry has emerged as a powerful tool for unbiased, comprehensive, and quantitative investigation of histones in clinical samples, with recent advancements in technology expanding its application possibilities. The 'bottom-up' strategy of analyzing short proteolytic peptides has shown success in clinical sample analysis, offering a promising approach for histone PTM profiling.

aniFOUND: analysing the associated proteome and genomic landscape of the repaired nascent non-replicative chromatin

Summary: The study developed aniFOUND, an antibody-free method for labeling, capturing, mapping, and characterizing nascent chromatin fragments synthesized in response to specific cues. Mass-spectrometry identified factors with unknown roles in UVC-DNA damage response and validated repair-dependent recruitment of chromatin remodeller and cohesin-loader proteins at specific photolesion sites. Developing aniFOUND-seq allowed high-resolution mapping of UDS activity for monitoring UVC repair-synthesis events genome wide.
