The existence of Aβ strains and their potential for driving phenotypic heterogeneity in Alzheimer's disease

Heather H. C. Lau et al.

Summary: Alzheimer's disease shares considerable clinical and pathological variability with human prion diseases. A beta peptide can assemble into a spectrum of aggregate strains similar to prion protein, exhibiting key hallmarks of prion strains and potentially driving phenotypic heterogeneity in Alzheimer's disease.

Mutant Huntingtin Is Cleared from the Brain via Active Mechanisms in Huntington Disease

Nicholas S. Caron et al.

Summary: This study investigated the mechanisms of mHTT clearance from the brain in adult mice to elucidate the significance of therapy-induced CSF mHTT changes. The data supports both passive release and active clearance of mHTT into CSF, suggesting that its treatment-induced changes may represent a combination of target engagement and preservation of neurons.

Ageing promotes pathological alpha-synuclein propagation and autonomic dysfunction in wild-type rats

Nathalie Van den Berge et al.

Summary: Using a rat model, it was found that the propagation of alpha-synuclein pathology from the gut to the brain is more efficient in old rats, resulting in dysfunction of the heart and stomach similar to observations in patients with Parkinson's disease. Age was shown to be a crucial factor for the aggregation and propagation of alpha-synuclein, highlighting the importance of using aged animals in preclinical studies for better understanding of age-dependent interactions.

Small, Seeding-Competent Huntingtin Fibrils Are Prominent Aggregate Species in Brains of zQ175 Huntington's Disease Knock-in Mice

Franziska Schindler et al.

Summary: The study demonstrates the presence of seeding-competent, fibrillar mHTT aggregates in the brains of zQ175 knock-in HD mice. These findings suggest that small, fibrillar, seeding-competent mHTT structures are prominent aggregate species in the brains of zQ175 mice.

The Contribution of Somatic Expansion of the CAG Repeat to Symptomatic Development in Huntington's Disease: A Historical Perspective

Darren G. Monckton

Summary: The discovery of unstable simple sequence repeats as the causative mutation for inherited human disorders such as Huntington's disease in the early 1990s opened up new insights into the genetic mechanisms underlying these diseases. Understanding the unstable nature of these repeats in both germline and somatic cells has shed light on the wide symptomatic variability observed in Huntington's disease and related disorders. These insights have also provided new avenues for potential therapies by targeting the somatic expansion of these repeats and identifying novel protein targets for intervention. Exciting prospects lie ahead in translating these findings into novel treatments for Huntington's disease and related disorders.
