Quantitative Chemical Profiling of Commercial Glyceride Excipients via1H NMR Spectroscopy

Isha Saraf et al.

Summary: This study aimed to develop a calibration-free quantitative proton nuclear magnetic resonance (qHNMR) method for the simultaneous quantification of different components of glycerides. The accuracy, precision, and robustness of the method were evaluated and found suitable for quantitative analysis of marketed excipients. The method demonstrated potential for identifying variability in glycerides among different batches and suppliers, as well as discerning changes during storage.

Interplay of Aging and Lot-to-Lot Variability on the Physical and Chemical Properties of Excipients: A Case Study of Mono- and Diglycerides

Varun Kushwah et al.

Summary: This study investigates the influence of chemical composition on the physical properties of mono- and diglycerides (MDGs) at a microstructural level, particularly in relation to aging and lot-to-lot variability. The findings suggest that changes in the chemical composition of MDG, potentially due to aging and lot-to-lot variability, can impact the physical structures and stability of emulsions. The research provides insights into the interdependency of the physicochemical properties of MDG and can serve as a guidance for establishing specifications tailored to different applications.

Liquid Lipids Act as Polymorphic Modifiers of Tristearin-Based Formulations Produced by Melting Technologies

Serena Bertoni et al.

Summary: This study evaluated the impact of different oral-approved liquid lipids on the polymorphism, phase transitions, and structure of solid lipid-based formulations, and their influence on drug release. The addition of liquid lipids was found to promote the transition of tristearin from the alpha-form to the stable beta-form, affecting the crystal structure of tristearin-based formulations and influencing the release behavior of a model hydrophilic drug. Overall, the addition of liquid lipids can be considered an interesting approach to control triglyceride crystallization in the beta-form, enabling the production of stable lipid formulations.
