Principles of nucleosome recognition by chromatin factors and enzymes

Robert K. McGinty et al.

Summary: The recent cryoelectron microscopy structures of chromatin complexes show that many chromatin proteins interact with their nucleosome substrate by using similar arginine anchor and variant arginine interactions. These proteins target specific histone structures on the nucleosome surface and can also interact with and distort nucleosomal DNA for complementary binding.

Rfam 14: expanded coverage of metagenomic, viral and microRNA families

Ioanna Kalvari et al.

Summary: Rfam is a database of RNA families with 3444 families, each represented by a multiple sequence alignment of known RNA sequences and a covariance model for searching additional members. Recent developments focused on improving data quality and coverage, adding new families like microRNAs, viral and bacterial RNAs through expert collaborations. The database saw significant growth with 759 new families added in Rfam 14, along with new features such as the Rfam Cloud family curation system.

Short H2A histone variants are expressed in cancer

Guo-Liang Chew et al.

Summary: Short H2A variants are testis-specific histones that destabilize nucleosomes during spermatogenesis. This study demonstrates their expression in a variety of cancers and identifies splicing changes associated with nucleosome instability in these malignancies. The authors suggest that short H2As may function as a class of ready-made oncohistones contributing to chromatin dysfunction in cancer.

Histone variants at a glance

Paul B. Talbert et al.

Summary: Eukaryotic nucleosomes are organized by histones, with common histones deposited during S-phase and histone variants available throughout the cell cycle. Variants alter nucleosome structure and function, playing specialized roles in DNA repair, chromosome segregation, and transcriptional regulation.

The structure of a virus-encoded nucleosome

Marco Igor Valencia-Sanchez et al.

Summary: The cryo-EM structure of DNA-assembled histone pairs from Marseillevirus, a nucleocytoplasmic large DNA virus, reveals that they form viral nucleosomes with highly conserved features similar to canonical eukaryotic nucleosomes. Viral histones from Marseillevirus form 'forced' heterodimers that assemble into tetramers, which in turn assemble DNA to form structures virtually identical to canonical eukaryotic nucleosomes.

Virus-encoded histone doublets are essential and form nucleosome-like structures

Yang Liu et al.

Summary: Recent studies have challenged the traditional view of nucleosome organization in eukaryotes by identifying minimalist histones in archaea and discovering genes encoding remote homologs of eukaryotic histones in giant viruses. The presence of viral doublet histones, essential for viral infectivity, suggests a specialized function of histones in the biology of these unusual viruses. Cryo-EM structures of viral nucleosome-like particles show similarities to eukaryotic nucleosomes despite limited sequence identity, further expanding the range of organisms that require nucleosomes.

Histone variants take center stage in shaping the epigenome

Michael Borg et al.

Summary: The dynamic properties of the nucleosome are crucial to genomic activity, with histone variants playing a pivotal role in modulating nucleosome structure and function. Specifically in plant chromatin landscapes, deposition of replicative H3.1 controls epigenetic inheritance while local replacement of H3.1 with H3.3 marks cells undergoing terminal differentiation. Deposition of specialized H3 variants in specific cell types is emerging as a novel mechanism of selective epigenetic reprogramming during the plant life cycle.
