Orphan nuclear receptors as regulators of intratumoral androgen biosynthesis in castration-resistant prostate cancer

Jianfu Zhou et al.

Summary: CRPC typically occurs after ADT, driven by AR reactivation and upregulation of steroidogenic enzymes. ONRs play crucial roles in the transcriptional regulation and promotion of intratumoral androgen synthesis, potentially serving as therapeutic targets for CRPC.

Inhibition of Estrogen-Related Receptor α Blocks Liver Steatosis and Steatohepatitis and Attenuates Triglyceride Biosynthesis

Chien-Yu Chen et al.

Summary: Inhibiting ERR alpha can block the development of NAFLD, reduce lipid accumulation, and attenuate NASH. Glycerolipid synthesis was identified as an additional mechanism for ERR alpha-regulated NAFLD/NASH development, with glycerophosphate acyltransferase 4 identified as a novel transcriptional target of ERR alpha. These results establish ERR alpha as a potential target for NAFLD/NASH treatment and highlight novel signaling pathways regulated by ERR alpha.

Estrogen-related receptor a is involved in angiogenesis and skeletal muscle revascularization in hindlimb ischemia

Danesh H. Sopariwala et al.

Summary: ERR alpha is induced in skeletal muscle by hindlimb ischemia or hypoxia, and its overexpression can drive a switch in myofiber type and activate gene programs related to angiogenesis in muscle. This leads to increased baseline capillarity and arterioles, as well as improved perfusion recovery in ischemic muscle, suggesting ERR alpha as a potential target for treating PAD/CLI.

Comprehensive Analysis of ESRRA in Endometrial Cancer

Shufang Wang et al.

Summary: This study comprehensively analyzed the expression level, clinical value, and possible mechanisms of action of ESRRA in uteri corpus endometrial carcinoma (UCEC), revealing that ESRRA may serve as a potential diagnostic and therapeutic target for UCEC. The findings highlighted the role of ESRRA in regulating cellular metabolism processes and influencing the tumorigenesis and progression of UCEC. Furthermore, the study demonstrated the potential of altering the immune microenvironment through targeting ESRRA for the development of immune checkpoint therapy in UCEC.
