Pathogenic Impacts of Dysregulated Polycomb Repressive Complex Function in Hematological Malignancies

Satoshi Kaito et al.

Summary: Polycomb repressive complexes play a crucial role in maintaining hematopoietic stem cells and mutations in corresponding genes are associated with hematologic malignancies, leading to resistance to chemotherapy and poor prognosis. Recent studies have revealed that these gene mutations are druggable targets, and inhibitors have shown therapeutic efficacy for tumors.

Molecular biology of Hodgkin lymphoma

Marc A. Weniger et al.

Summary: Classical Hodgkin lymphoma (cHL) has unique biological features such as the orchestration of a complex microenvironment by HRS cells, the major role of Epstein-Barr virus in lymphomagenesis, and the defining interplay of genetic lesions and cellular origin. These factors contribute to the constitutive activation of multiple signaling pathways in cHL.

Bcor deficiency perturbs erythro-megakaryopoiesis and cooperates with Dnmt3a loss in acute erythroid leukemia onset in mice

Paolo Sportoletti et al.

Summary: Recurrent loss-of-function mutations of the BCOR gene are associated with poor prognosis in AML patients, leading to the creation of a mouse model to study this AML genotype. Double knockout mice showed features of acute erythroid leukemia, suggesting potential efficacy of demethylating agents in AEL treatment.
