Kinetic and Structural Characterization of the Self-Labeling Protein Tags HaloTag7, SNAP-tag, and CLIP-tag

Summary: The study compared the labeling kinetics and substrate specificities of self-labeling protein tags (SLPs) HaloTag7, SNAP-tag, and CLIP-tag. It was found that HaloTag7 reaches almost diffusion-limited labeling rate constants with some substrates, while SNAP-tag labeling rate constants are less affected by the structure of the label. Crystal structures of HaloTag7 and SNAP-tag labeled with fluorescent substrates helped explain their substrate preferences and aided in the design of substrates with improved labeling kinetics.
