Cubilin-, megalin-, and Dab2-dependent transcription revealed by CRISPR/Cas9 knockout in kidney proximal tubule cells

Summary: This study used RNA-sequencing and CRISPR/Cas9 knock-out technology to investigate the transcriptional changes in proximal tubule cells caused by the loss of megalin, cubilin, or Dab2 expression. The results showed that Lrp2 KO had the greatest effect on transcription, and the affected genes in Cubn KO and Dab2 KO cells were also changed in Lrp2 KO cells. Pathway analysis suggested changes in immunomodulatory pathways that may contribute to the observed pathological changes. Differences in transcription patterns between Lrp2 and Dab2 KO cells also suggested the involvement of aberrantly localized receptors in the transcriptional changes. This study highlights the role of megalin as a master regulator in proximal tubule cells.
