Detection and Characterization of Vesicular Gangliosides Binding to Myelin-Associated Glycoprotein on Supported Lipid Bilayers

Jennie L. Cawley et al.

Summary: The study reveals that ganglioside-MAG interactions between axons and myelin can inhibit neuronal growth, and presents a real-time method to characterize these interactions. By controlling the content of gangliosides and MAG, and presenting them in their in vivo orientation within fluid lipid bilayers, the study found the dynamics of MAG-ganglioside interactions.

A Systematic Study of Unsaturation in Lipid Nanoparticles Leads to Improved mRNA Transfection In Vivo

Sang M. Lee et al.

Summary: The study identified an ionizable lipid series (4A3) with a citronellol-based (Cit) periphery as the best performer out of 91 amino lipids, showing superior lipid fusion and improved mRNA delivery efficacy in vivo through Selective ORgan Targeting (SORT). These findings demonstrate how lipid unsaturation promotes mRNA delivery and how lipid mixing can enhance efficacy.

Heteromultivalent topology-matched nanostructures as potent and broad-spectrum influenza A virus inhibitors

Chuanxiong Nie et al.

Summary: This study demonstrates the topology-matched design of heteromultivalent nanostructures as potent and broad-spectrum virus entry inhibitors based on the host cell membrane. By transferring heteromultivalent binding moieties to nanostructures with a bowl-like shape matching the viral surface, the inhibitors show superior performance in virus propagation inhibition. With identified binding sites on the S protein of SARS-CoV-2, this approach holds promise in developing effective inhibitors to prevent coronavirus infection.

A Dynamic, Supramolecular View on the Multivalent Interaction between Influenza Virus and Host Cell

Nico J. Overeem et al.

Summary: This review explores how influenza viruses navigate mucus and recognize host cells through multivalent interactions. The surface proteins of the influenza A virus facilitate specific binding to host cells, and the importance of these multivalent interactions in crossing mucus and entering host cells is discussed. Techniques to measure the avidity and receptor specificity of influenza viruses are also reviewed.

Unraveling How Multivalency Triggers Shape Deformation of Sub-100 nm Lipid Vesicles

Hyeonjin Park et al.

Summary: This study tracked the attachment of sub-100 nm lipid vesicles to a streptavidin-functionalized supported lipid bilayer using LSPR sensing technique and developed an analytical model to extract quantitative details. The findings reveal how varying the surface densities of streptavidin receptors and biotin ligands affects nanometer-scale vesicle deformation, identifying a critical ligand density where appreciable vesicle deformation began. These results provide insight into the interplay between membrane bending energy and multivalent binding interaction energy in various multivalent ligand-receptor systems.

Quantitative Nanomechanical Analysis of Small Extracellular Vesicles for Tumor Malignancy Indication

Siyuan Ye et al.

Summary: In this study, the nanomechanical properties of human breast cancer cell-derived sEVs were quantitatively investigated using atomic force microscopy (AFM). It was found that the stiffness and osmotic pressure of sEVs increase with malignancy and decrease with size, while the bending modulus decreases with malignancy and is lower in smaller sEVs. This research establishes a relationship between the nanomechanical signature of sEVs and tumor malignancy, contributing to a better understanding of cancer mechanobiology.
