Alteration of circRNA and lncRNA expression profile in exosomes derived from periodontal ligament stem cells undergoing osteogenic differentiation

Summary: This study investigated the expression profile of circRNAs and lncRNAs in exosomes derived from periodontal ligament stem cells before and after osteogenic differentiation, revealing significant changes in the early phase of differentiation related to pathways regulating stem cell pluripotency. Further studies would be needed to explore the roles of exosomal circRNAs and lncRNAs in osteogenic differentiation of PDLSCs.

Summary: This study presents a protocol for isolating multiple EV subpopulations directly from tissues, including enzymatic treatment, ultracentrifugation, density separation, electron microscopy characterization, etc., which can be completed in 22 hours. The approach can be applied to tissues such as human melanoma metastases and can be adopted by researchers experienced with cell culture and EV isolation.

Salivary Outer Membrane Vesicles and DNA Methylation of Small Extracellular Vesicles as Biomarkers for Periodontal Status: A Pilot Study

Summary: The study compared salivary sEVs from periodontitis patients, healthy controls, and gingivitis patients, finding that global 5mC hypermethylation in salivary sEVs can serve as a potential biomarker with high sensitivity and specificity for distinguishing periodontitis patients. Periodontitis sEVs showed significantly increased levels of LPS+ OMVs, global 5mC methylation, and periodontal pathogens, which were more pronounced in sEVs than whole saliva.

Metabolomics of gingival crevicular fluid to identify biomarkers for periodontitis: A systematic review with meta-analysis

The results showed differential expression of certain metabolites in GCF across different periodontal conditions, highlighting their potential for investigating periodontal pathophysiology and site-specific diagnosis. Oxidative stress-related molecules, particularly malondialdehyde and 8-hydroxy-deoxyguanosine, were consistently associated with periodontitis.
