Triazolyl-Functionalized N-Heterocyclic Carbene Half-Sandwich Compounds: Coordination Mode, Reactivity and in vitro Anticancer Activity

Kelvin K. H. Tong et al.

Summary: Investigations into the anticancer activity of organometallic complexes of N-heterocyclic carbenes with a triazolyl moiety reveal potential for cytotoxicity, with the nature of the metal center and ligand lipophilicity playing significant roles in determining biological properties. The Ir-III complex 5 d showed the highest cytotoxicity against NCI-H460 non-small cell lung carcinoma cells, with localization in the cytoplasm observed through cell uptake and distribution studies using X-ray fluorescence microscopy.

The interaction of rhodium compounds with proteins: A structural overview

Domenico Loreto et al.

Summary: Rhodium compounds have been investigated for their interactions with peptides and proteins to create artificial metalloenzymes with enhanced catalytic features. The formation of Rh/peptides and Rh/protein adducts and the structures of these complexes have been analyzed, providing implications for the design of Rh-based artificial metalloenzymes. Covalent and dative anchoring strategies can be used to attach Rh compounds to proteins, with Rh centers preferentially binding specific amino acid side chains.

Anthracenyl Functionalization of Half-Sandwich Carbene Complexes: In Vitro Anticancer Activity and Reactions with Biomolecules

Betty Y. T. Lee et al.

Summary: N-Heterocyclic carbene (NHC) ligands have been widely studied in medicinal inorganic chemistry. In this study, a series of half-sandwich [M(L)(NHC)Cl-2] complexes with N-flanking anthracenyl moiety were prepared and their anticancer activities were investigated, showing cytotoxicity in low micromolar range. The Ru derivative 1a exhibited stability and specific interactions with biomolecules.

Mitochondria-targeted cyclometalated rhodium(iii) complexes: synthesis, characterization and anticancer research

Yan-Bo Peng et al.

Summary: Inorganic medicinal chemistry has largely focused on platinum or ruthenium, with insufficient research on other metal centers such as rhodium. This study synthesized a series of cyclometalated rhodium(iii) complexes, which showed stronger anticancer activity than cisplatin. Mechanistic studies revealed that these complexes induced cellular apoptosis through mitochondrial pathways by causing severe damage and cell cycle arrest.

Rhodium(i) N-heterocyclic carbene complexes: synthesis and cytotoxic properties

Ichraf Slimani et al.

Summary: A series of rhodium(i) complexes with N-heterocyclic carbene ligands showed high in vitro cytotoxic activity against human cancer cells, with complex 2b exhibiting the most selective cytotoxic activity. These complexes demonstrated greater cytotoxicity against cancer cells compared to established anticancer agents cisplatin and 5-fluorouracil.
