Summary: A base-controlled protocol was developed for the C-N coupling of primary amines and 2-chlorobenzimidazoles, achieving selective synthesis of secondary or tertiary amines. The selectivity can be switched from monoarylation to diarylation by changing the base from iPr(2)NH to LiOtBu. The type, intrinsic basicity, and amount of bases utilized greatly affect the reactions.

Transition-Metal-Free and Base-Promoted Carbon-Heteroatom Bond Formation via C-N Cleavage of Benzyl Ammonium Salts

Summary: A facile and general method for constructing carbon-heteroatom bonds via C-N cleavage of benzyl ammonium salts under transition-metal-free conditions was reported. The combination of t-BuOK and 18-crown-6 enabled the coupling of a wide range of substituted benzyl ammonium salts with heteroatom nucleophiles. Good functional group tolerance was demonstrated, and scale-up reactions and one-pot syntheses were successfully performed.

Base-Promoted Tandem Synthesis of 2-Azaaryl Tetrahydroquinolines

Summary: A novel method for synthesizing 2-azaaryl tetrahydroquinolines is reported by the base-promoted tandem reaction of azaaryl methyl amines and styrene derivatives. Mechanistic probe experiments confirm the mechanism involving deprotonation of the benzylic C-H bond and addition to the styrene vinyl group via the SNAr mechanism. High yields of up to 95% were achieved in over 30 examples, demonstrating the potential applicability of this method.

Switchable and Scalable Heteroarylation of Primary Amines with 2-Chlorobenzothiazoles under Transition-Metal-Free and Solvent-Free Conditions

Summary: A new switchable and scalable C-N coupling protocol was developed for the efficient synthesis of 2-aminobenzothiazoles, without the need for transition metals and solvents. The addition of an appropriate amount of NaH could completely switch the selectivity and increase the yield of the products, allowing for gram-scale synthesis with simple procedures. Evaluation of green metrics showed that the protocol was superior to existing methods in terms of green chemistry and sustainability.

A practical and sustainable protocol for direct amidation of unactivated esters under transition-metal-free and solvent-free conditions

Summary: A NaOtBu-mediated synthesis approach was developed for direct amidation of unactivated esters with amines under transition-metal-free and solvent-free conditions, affording a series of amides in good to excellent yields at room temperature. This environmentally friendly and practical method can be applied for the synthesis of valuable molecules and has the potential to streamline amide synthesis in industry. The green metrics evaluation of all the target products indicates the advantages of this protocol in terms of environmental friendliness and sustainability.
