Enhanced Akt/GSK-3β/CREB signaling mediates the anti-inflammatory actions of mGluR5 positive allosteric modulators in microglia and following traumatic brain injury in male mice

Shahnawaz A. Bhat et al.

Summary: Research shows that mGluR5 PAM treatment post-TBI reduces pro-inflammatory microglial activation by modulating Akt/GSK-3 beta/CREB signaling, enhancing anti-inflammatory markers and promoting neuroprotection.

Conditional genetic deletion of CSF1 receptor in microglia ameliorates the physiopathology of Alzheimer's disease

Vincent Pons et al.

Summary: The study showed that CSF1R KO did not impair microglial survival and proliferation in 6- and 8-month-old mice. Cognitive decline was delayed in CSF1R-deleted mice, associated with a decrease in plaque volume. Following the knockout, a compensatory mechanism involving increased TREM2/beta-Catenin and IL-34 expression may promote microglial survival and A beta phagocytosis.

INPP5D expression is associated with risk for Alzheimer's disease and induced by plaque-associated microglia

Andy P. Tsai et al.

Summary: Alzheimer's disease (AD) is a progressive neurodegenerative disorder with microglia playing a prominent role in late-onset AD. Studies have shown that inositol polyphosphate-5-phosphatase (INPP5D) is selectively expressed in brain microglia and its increased expression may be a risk factor in AD. INPP5D expression increases as the disease progresses, particularly in plaque-associated microglia, suggesting INPP5D as a potential therapeutic target for AD.

Activated microglia mitigate Aβ-associated tau seeding and spreading

Maud Gratuze et al.

Summary: Studies have shown that TREM2 deficiency and microglial ablation significantly increase tau seeding and spreading, and while repopulated microglia cluster around plaques, they have reduced disease-related gene expression and increased tau seeding/spreading.
