Summary: By analyzing the transcriptome landscape in human adipocytes, this study revealed elevated mitochondrial ROS pathway and NF-KB signaling, as well as altered fatty acid metabolism in T2DM adipocytes. Mouse experiments showed that adipose-specific deletion of mitochondrial Trx2 led to excessive mitophagy, increased inflammation, and lipolysis in white adipose tissues. Treatment with ROS scavenger or NF-KB inhibitor improved metabolic disorders and T2DM progression in mice.

Understanding the role of nucleic acids in obesity-induced inflammation may lead to the development of new therapeutic approaches to mitigate obesity-related complications.

High extracellular ATP levels released through pannexin-1 channels mediate inflammation and insulin resistance in skeletal muscle fibres of diet-induced obese mice

Summary: In obese mice, skeletal muscle fibres release high levels of extracellular ATP through overexpressed PANX1 channels, leading to an inflammatory state and insulin resistance. The reduction of extracellular ATP levels by PANX1 blockers or knockdown improves inflammation and insulin sensitivity in skeletal muscle fibres. Incubation with PANX1 blockers rescues insulin resistance and improves glycaemic management in obese mice.
