Chip-calorimetric assessment of heat generation during Ca2+ uptake by digitonin-permeabilized Trypanosoma cruzi

Marina Rincon Sartori et al.

Summary: Ca2+ signaling in trypanosomatids plays an important role in energy metabolism regulation, with Ca2+ concentration being finely regulated within cellular compartments. Research shows that heat is generated during Ca2+ uptake by mitochondria and acidocalcisomes, with mitochondria exhibiting minimal heat dissipation and ATPase pumps in acidocalcisomes contributing significantly to the heat generated.

The mitochondrial F1FO-ATPase exploits the dithiol redox state to modulate the permeability transition pore

Cristina Algieri et al.

Summary: The dithiol reagents phenylarsine oxide (PAO) and dibromobimane (DBrB) have opposite effects on the F1FO-ATPase activity, with PAO increasing the activity and DBrB decreasing it. Additionally, PAO stimulates the formation of the mitochondrial permeability transition pore (mPTP) while DBrB desensitizes it.

The Mitochondrial Permeability Transition: Nexus of Aging, Disease and Longevity

Hagai Rottenberg et al.

Summary: mPTP activity is enhanced in aging and aging-driven degenerative diseases, releasing harmful substances that accelerate cell aging and disease progression. Autophagy, a protective mechanism, depends on the inhibition of mPTP for its effectiveness. Inhibition of autophagy and mitophagy in aging cells leads to increased mPTP activation, contributing to neurodegenerative diseases. Enhancing autophagy and inhibiting mPTP activation through drugs and lifestyle modifications can promote longevity and healthspan.

Feeding effects on liver mitochondrial bioenergetics of Boa constrictor (Serpentes: Boidae)

Helena Rachel da Mota Araujo et al.

Summary: Snakes, as interesting examples of taxa that can overcome energy metabolism challenges, exhibit dual extreme adaptations. The study on Boa constrictor snakes showed that liver mitochondria increase respiration rates during the postprandial period, improving bioenergetic capacity without compromising redox balance. Despite the increase in H2O2 release, no differences were found in susceptibility to mitochondrial permeability transition or mitochondrial redox state between fasted and fed animals.

Modulation and Pharmacology of the Mitochondrial Permeability Transition: A Journey from F-ATP Synthase to ANT

Andrea Carrer et al.

Summary: The permeability transition (PT) involves increased permeation of the inner mitochondrial membrane due to the opening of the PT pore (PTP), which is a Ca2+-activated high conductance channel. Despite extensive exploration on PTP modulation, the lack of a clear understanding of its molecular nature complicates any target-based approach. Recent advancements indicate the existence of at least two mitochondrial permeability pathways mediated by the F-ATP synthase and the ANT, but the exact molecular mechanism leading to channel formation remains elusive for both.

Defining the molecular mechanisms of the mitochondrial permeability transition through genetic manipulation of F-ATP synthase

Andrea Carrer et al.

Summary: The nature of the mitochondrial permeability transition pore (PTP) remains debated, with the possibility that it can be mediated by F-ATP synthase or adenine nucleotide translocator. Through genetic modifications and cell experiments, researchers identified the essential role of certain mitochondrial subunits in turning the F-ATP synthase into the PTP, and showed that the ANT can contribute to channel formation in the absence of an assembled F-ATP synthase.
